Recent Advances in RP-HPLC Method Development and Validation for Empagliflozin: A Comprehensive Review

Main Article Content

Abstract

Empagliflozin, a selective sodium-glucose co-transporter 2 (SGLT2) inhibitor, is widely prescribed for the treatment of type 2 diabetes mellitus, either alone or in fixed-dose combinations. Accurate and robust analytical methods are essential to ensure its quality, safety, and efficacy in bulk drug and pharmaceutical dosage forms. Among various techniques, reversed-phase high-performance liquid chromatography (RP-HPLC) remains the most widely adopted due to its versatility, sensitivity, and regulatory acceptance. This review provides a comprehensive overview of the development and validation of RP-HPLC methods for the estimation of Empagliflozin. Key physicochemical properties such as pKa, solubility, and UV absorbance characteristics are discussed in the context of chromatographic behavior. The article also outlines critical regulatory requirements from ICH Q2(R1/R2), pharmacopeial standards, and method validation parameters including specificity, linearity, precision, accuracy, robustness, and stability.A survey of published RP-HPLC methods is presented, covering both mono-component and combination formulations with Metformin, Linagliptin, and Sitagliptin. Comparative analysis of chromatographic conditions—such as mobile phase composition, detection wavelength, column type, and system suitability—highlights best practices and common challenges. The article further explores method sensitivity (LOD/LOQ), forced degradation studies, and the need for stability-indicating assays.

Future directions emphasize the adoption of green solvents, UHPLC technologies, Analytical Quality by Design (AQbD), and harmonization of analytical protocols. Recommendations are provided for bioanalytical extensions, improved impurity profiling, and establishment of standard or platform methods. This review serves as a valuable resource for researchers and analysts engaged in the pharmaceutical quality control and regulatory evaluation of Empagliflozin.
