Abstract

Objective of present investigation was to develop carvedilol phosphate floating microsphere for retention in the upper part of the GIT to improve the dissolution where the solubility of carvedilol phosphate is more in acidic medium. Floating Microspheres were prepared by emulsion solvent diffusion method using ethylcellulose and eudragit RS 100 as polymer and 0.5% w/v polyvinyl alcohol as external medium. 32 full factorial design was used for optimizing the drug: polymer ratio(X1) and ethylcellulose: eudragit RS 100(X2) ratio were selected as independent variables, evaluated for various parameters which were analysed using ANOVA and surface response methodology. Batch F5 was selected as optimized, as provides desired zero order release profile as well as 77.25% buoyancy and 80.66% entrapment efficiency, 82.9% yield and mean particle size of 133.86 μm and spherical microsphere obtained confirmed by SEM study. Drug and excipients are compactible to each other was confirmed by FTIR study.
